Pine, Leo (Communicable Disease Center, Atlanta, Ga.), Clarence J. Boone, and Dave McLaughlin. Antigenic properties of the cell wall and other fractions of the yeast form of Histoplasma capsulatum. J. Bacteriol. 91:2158-2168. 1966.-Yeast-form cells of Histoplasma capsulatum were fractionated in an attempt to obtain complement-fixing antigens with greater specificity than those of the currently used whole yeast form. No specific fraction was isolated. Rupture of the whole cell produced soluble and insoluble highly antigenic fractions. The soluble fractions rapidly lost antigenicity. Marked reduction of activity of some particulate fractions was also observed. The purified cell wall, stripped of its protein components, was antigenic, relatively stable, and demonstrated low cross-reactivity with heterologous sera. All fractions cross-reacted with sera from cases of North American blastomycosis to a greater degree than did the whole yeast-form antigen of H. capsulatum. The production of certain cellular fractions varied with the strains used. In complement-fixation tests, insignificant differences in specificity and antigenicity were found with whole-cell antigens from 13 H. capsulatum strains as well as with purified cell wall fractions prepared from 2 of these strains.

Recognition of Histoplasma capsulatum yeast-cell antigens by human lymphocytes and human T-cell clones.
We have prepared a detergent extract from the cell wall and cell membrane of Histoplasma capsulatum yeast cells that is recognized by T cells from mice immunized with viable organisms or with the extract. A 62-kd antigen from this extract has been isolated and has been shown to be antigenic and to confer protective immunity in mice. In this study, we examined the in vitro proliferative response...
متن کاملCranial and Spinal Locations of Histoplasma capsulatum var. duboisii in Brazzaville
Background: Histoplasma capsulatum var. duboisii is a rare fungus, endemic in southern Africa of the Sahara and Madagascar. Objective: To describe the confirmed cases of histoplasmosis. Methods: This retrospective study was conducted at the division of neurosurgery of Brazzaville academic hospital in the Republic of Congo. Clinical records of all confirmed cases who were admitted between Jan...
متن کاملPhenotypic variation and intracellular parasitism by histoplasma Capsulatum.
The success of Histoplasma capsulatum as an intracellular pathogen depends completely on successful conversion of the saprophytic mycelial (mold) form of this fungus to a parasitic yeast form. It is therefore not surprising that yeast phase-specific genes and gene products are proving to be important for survival and proliferation of H. capsulatum within macrophages. In this study, we have focu...
متن کاملThe interaction between Histoplasma capsulatum cell wall carbohydrates and host components: relevance in the immunomodulatory role of histoplasmosis.
Histoplasma capsulatum is an intracellular fungal pathogen that causes respiratory and systemic disease by proliferating within phagocytic cells. The binding of H. capsulatum to phagocytes may be mediated by the pathogen's cell wall carbohydrates, glucans, which consist of glucose homo and hetero-polymers and whose glycosydic linkage types differ between the yeast and mycelial phases. The alpha...
متن کاملMonosaccharide and chitin content of cell walls of Histoplasma capsulatum and Blastomyces dermatitidis.
Cell walls of Histoplasma capsulatum and Blastomyces dermatitidis, obtained by mechanical breakage of yeast- and mycelial-phase cultures, were lipid-extracted and then fractionated with ethylenediamine. Unextracted cell walls, lipid-extracted cell walls, and the three fractions resulting from ethylenediamine treatment were examined for monosaccharide and chitin content. The yeast-phase cell wal...
